IL-2 is a powerful immunoregulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. It is expressed by CD4+ and CD8+ T cells, gammadelta T cells, B cells, dendritic cells, and eosinophils. IL-2/IL-2R signaling is required for T-cell proliferation and other fundamental functions which are essential for the immune response. The receptor for IL-2 consists of three subunits (55 kDa IL2Ralpha, 75 kDa IL2Rβ, 64 kDa common gamma chain gammac/IL2Rgamma) that are present on the cell surface in varying preformed complexes. Mature human IL-2 shares 56 % and 66 % amino acid sequence identity with mouse and rat IL-2, respectively. Human and mouse IL-2 exhibit cross-species activity.

Interleukin-1 alpha (IL-1alpha) is a non-secreted proinflammatory cytokine produced mainly by activated macrophages, as well as neutrophils, epithelial cells, and endothelial cells. It possesses metabolic, physiological, haematopoietic activities, and plays one of the central roles in the regulation of the immune responses. Both IL-1alpha and IL-1β binds to the same receptor and has similar identical biological properties. Among various species, the amino acid sequence of mature IL-1alpha is conserved 60 % to 70 % and human IL-1 has been found to be biologically active on murine cell lines. IL-1alpha recently started to find effective application in cosmetic and dermatological formulations, which allow to significantly harmonizing derma architecture.

It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. Interleukin-10 (IL-10), also known as cytokine synthesis inhibitory factor (CSIF), is the charter member of the IL-10 family of α-helical cytokines that also includes IL-19, IL-20, IL-22, IL-24, and IL-26/AK155. IL-10 is secreted by many activated hematopoietic cell types as well as hepatic stellate cells, keratinocytes, and placental cytotrophoblasts. Whereas human IL-10 is active on mouse cells, mouse IL-10 does not act on human cells. IL-10 is a 178 amino acid molecule that contains two intrachain disulfide bridges and is expressed as a 36 kDa noncovalently associated homodimer. The IL-10 dimer binds to two IL-10 Rα/IL-10R1 chains, resulting in recruitment of two IL-10 Rβ/IL-10R2 chains and activation of a signaling cascade involving JAK1, TYK2, and STAT3. IL-10Rβ does not bind IL-10 by itself but is required for signal transduction. IL-10 is a critical molecule in the control of viral infections and allergic and autoimmune inflammation. It promotes phagocytic uptake and Th2 responses but suppresses antigen presentation and Th1 proinflammatory responses.

IL-12 is a heterodimeric cytokine of disulfide-linked p35 and p40 subunits. It is secreted by monocytes, macrophages, dendritic cells, neutrophils, Langerhans cells, keratinocytes, microglia, and peripheral B cells. In humans, the p40 subunit can also associate with a p19 subunit to form IL-23. IL-12 signals through a receptor complex consisting of IL-12 R beta 1 and IL-12 R beta 2. It promotes Th1 immune responses by inducing IFN-gamma secretion from NK cells, T cells, and macrophages and by enhancing NK cell and T cell mediated cytotoxicity. It also cooperates with IL-18 in the generation of Th1 memory cells.
