Interferon-gamma (IFN-gamma), also known as Type II interferon or immune interferon, is a cytokine produced primarily by T-lymphocytes and natural killer cells. The protein shares no significant homology with IFN-β or the various IFN-alpha family proteins. Mature IFN-gamma exists as noncovalently-linked homodimers. Human IFN-gamma is highly species specific and is biologically active only in human and primate cells. IFN-gamma was originally characterized based on its antiviral activities. The protein also exerts antiproliferative, immunoregulatory and proinflammatory activities and is thus important in host defense mechanisms. IFN-gamma induces the production of cytokines, upregulates the expression of class I and II MHC antigens, Fc receptor and leukocyte adhesion molecules. It modulates macrophage effector functions, influences isotype switching and potentiates the secretion of immunoglobulins by B cells. IFN-gamma also augments TH1 cell expansion and may be required for TH1 cell differentiation.

Epidermal Growth Factor (EGF) was originally discovered in crude preparations of nerve growth factor prepared from mouse submaxillary glands as an activity that induced early eyelid opening, incisor eruption, hair growth inhibition, and stunting of growth when injected into newborn mice. Human EGF was isolated from urine based on its inhibitory effect on gastric secretion and named urogastrone, accordingly. EGF is prototypic of a family of growth factors that are derived from membrane-anchored precursors. All members of this family are characterized by the presence of at least one EGF structural unit (defined by the presence of a conserved 6 cysteine motif that forms three disulfide bonds) in their extracellular domain. EGF is initially synthesized as a 130 kDa precursor transmembrane protein containing 9 EGF units. The mature soluble EGF sequence corresponds to the EGF unit located proximal to the transmembrane domain. The membrane EGF precursor is capable of binding to the EGF receptor and was reported to be biologically active. Mature human EGF shares 70 % a.a. sequence identity with mature mouse and rat EGF.

The insulin-like growth factors (IGFs) belonged to the insulin gene family, are mitogenic polypeptide growth factors that stimulate the proliferation and survival of various cell types including muscle, bone, and cartilage tissue in vitro. The IGFs are similar by structure and function to insulin, but have a much higher growth-promoting activity than insulin. IGF-1 is produced primarily by the liver as an endocrine hormone as well as in target tissues in a paracrine/autocrine fashion. The production of IGF-1 is stimulated by growth hormone (GH) and can be retarded by undernutrition, growth hormone insensitivity, lack of growth hormone receptors, or failures of the downstream signaling pathway post GH receptor including SHP2 and STAT5B. Recombinant human IGF-1 are globular proteins containing 70 amino acids and 3 intra-molecular disulfide bonds. Mature human IGF-1 shares 94 % and 96 % a.a. sequence identity with mouse and rat IGF-1, respectively, and exhibits cross-species activity.

Human bFGF, encoded by the FGF2 gene, is a member of the fibroblast growth factor (FGF) family. Fibroblast growth factor was found in pituitary extracts in 1973 and then tested in a bioassay that caused fibroblasts to proliferate. After further fractionating the extract using acidic and basic pH, two different forms have isolated that named “acidic fibroblast growth factor” (FGF-1) and “basic fibroblast growth factor” (FGF-2). Human bFGF shares 54 % amino acid sequence identity with aFGF. Affinity between bFGF and its receptors can be increased by heparin or heparan sulfate proteoglycan. bFGF plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. bFGF are also involved in a variety of biological processes, including embryonic development , morphogenesis, tissue repair, tumor growth and invasion. Additionally, bFGF is frequently used for a critical component of cell culture medium, e.g., human embryonic stem cell culture medium, serum-free culture systems.

Flt3-ligand (FL) is a recently identified hematopoietic cytokine whose activities are mediated by binding to the transmembrane glycoprotein Flt3. Flt3 was first discovered as a member of the class III subfamily of receptor tyrosine kinases (RTK) whose expression among hematopoietic cells was found to be restricted to highly enriched stem/progenitor cell populations. Additionally, class III RTKs include the receptors from SCF, M-CSF and PDGF. Not surprisingly, Flt3-ligand is also structurally related to M-CSF and SCF. All three cytokines have been shown to exist both as type I transmembrane proteins and as soluble proteins. The predominant human FL isoform is a transmembrane protein that can undergo proteolytic cleavage to generate a soluble form of the protein. An alternatively-spliced FL mRNA, encoding a soluble form of the human FL, has also been identified. FL is widely expressed in various human and mouse tissues. At the amino acid sequence level, human and mouse FL are approximately 72 % identical and the two proteins exhibit cross-species activity. FL has been shown to synergize with a wide variety of hematopoietic cytokines to stimulate the growth and differentiation of early hematopoietic progenitors.

Tumor necrosis factor alpha (TNF-alpha), also called cachectin, is the best-know member of the TNF-family, which can cause cell death. This protein is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-alpha occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-alpha is glycosylated, but non-glycosylated recombinant TNF-alpha has comparable biological activity. The biologically active native form of TNF-alpha is reportedly a trimer. Human and murine TNF-alpha show approximately 79 % homology at the amino acid level and cross-reactivity between the two species. Two types of receptors for TNF-alpha have been described and virtually all cell types studied show the presence of one or both of these receptor types.

Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF) is secreted by a number of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts) in response to cytokine or immune and inflammatory stimulation. It was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors and has functions of stimulates the growth and differentiation of hematopoietic precursor cells from various lineages. GM-CSF has also been reported to have a functional role on non-hematopoietic cells and can induce human endothelial cells to migrate and proliferate. Additionally, it can stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. Human GM-CSF shares 54 % sequences identity with mouse GM-CSF, but has no biological effects across species. GM-CSF is used as a medication to stimulate the production of white blood cells following chemotherapy and has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients.

Human Interleukin-15 (IL-15) is expressed by the IL15 gene located on the chromosome 4. It shares approximately 97 % and 73 % sequence identity with simian and murine IL-15, respectively. Both human and simian IL-15 are active on murine cells. IL-15 is secreted by mononuclear phagocytes (and some other cells), especially macrophages following infection by virus. It possesses a variety of biological functions, including stimulating and maintaining of cellular immune responses, especially regulating T and natural killer (NK) cell activation and proliferation. In additionally, it shares many biological properties with IL-2, including T, B and NK cell-stimulatory activities. IL-15 signals through a complex composed of IL-2/IL-15 receptor beta chain. Although IL-15 lacks sequence homology with IL-2, it has recently been shown that both the beta and gamma chains of the IL-2 receptor are utilized for IL-15 binding and signaling. In addition, an IL-15 specific binding protein has also been cloned from a mouse T cell clone.
